Background

Dry, thin, and sagging skin is a prevalent concern among both women and men aged 50 and above. This condition often results from the natural aging process, characterized by a decrease in the synthesis of vital lipids in the skin. Specifically, the decline in ω-O-acylceramides (acylCers), contributes to the manifestation of aged skin. AcylCers are crucial sphingolipids with a long acyl chain, usually between 30-36 carbons, and are essential for maintaining the barrier function of the epidermis thereby preventing uncontrolled water loss from the body. Deficiencies in acylCers extend beyond mere aging and are especially observed in certain severe skin disorders, such as psoriasis, atopic dermatitis, or ichthyosis. However, they are currently not available on an industrial scale and the enzymatic pathway to their biosynthesis was only recently discovered.
